The external polysaccharide is a major component of Micrococcus lysodeikticus cell wall and displays distinct composition. The complete structure of the external polysaccharide had been elucidated as a basis for investigation of the cell wall structure-function relation. However, the mode of attachment of the polysaccharide to the peptidoglycan through a phosphodiester was not clear due to limitations in structural and biosynthetic studies. The present study describes purification of a lysozyme-resistant nondialyzable high-molecular-weight fragment of cell wall and identifies the sugar, D-glucose, as the point of external polysaccharide attachment to the peptidoglycan through a phosphate diester. Kinetic studies for the acid-catalyzed release of external polysaccharide from the peptidoglycan were performed in parallel with synthetic [methyl-2-acetamido-3-O-(D-1-carboxyethyl)-2-deoxy-alpha-D- glucopyranoside-6-yl]-alpha-D-glucopyranosyl phosphate and alpha-D-glucopyranosyl phosphate and showed the presence of a phosphodiester linkage between external polysaccharide and peptidoglycan. In addition, type of phosphate residue and cross-linking between muramic acid and protein part have been determined.  相似文献

Bovine heart mitochondria suspended in 0.25 M sucrose were treated with 0.3% glutaraldehyde (GA). The membranes were disintegrated by ultrasonication in 0.25 M KCl and precipitated by centrifugation. The supernatant was assayed for creatine kinase (CKm) oligomeric forms by ultracentrifugation in a sucrose density gradient. A kinetic analysis of membrane-bound CKm was performed before and after ultrasonication. The data obtained suggest that the CKm octamer is the only form of CKm bound to mitochondrial membranes during GA treatment. This finding was confirmed by an analysis of extracts from untreated mitochondria using high resolution gel filtration.  相似文献

Raman spectra of eubacterial ribosomal 5S RNAs of Escherichia coli, Bacillus subtilis and Thermus thermophilis and of eukaryotic 5S RNAs of yeast and rat liver have been compared. The spectra show a very high and comparable regularity in the ribophosphate backbone as indicated by the ratio 1.67±0.03 for I812/I1100 in all samples. The 5S RNAs studied have a similar degree of stacking of the G, A and pyrimidine bases. A high percentage of base-paired U residues between 43 and 66% is indicated. Conformational alterations occurring in 5S RNAs in the presence of Mg2+ ions between 20 and 50*C are localized mainly in the region of loop II of the molecule. The implications of these results for the 5S RNA structure are discussed.  相似文献

It has been found that lymphocytes of hypercholesterolemic (HCh) subjects are characterized by a reduced number of glucocorticoid receptors (GcR) as compared with the cells of normolipidemics (N). Addition of HCh-sera or very low density lipoproteins, or low density lipoproteins isolated both from HCh-sera and N-sera to cultured human skin fibroblasts brought about a fall in the number of GcR in the cells. High density lipoproteins had no effect on GcR level. Dexamethasone was less effective in inhibiting cholesterol synthesis from [14C]acetate in the lymphocytes and fibroblasts with a reduced number of GcR. In the presence of dexamethasone (I x 10(-8)M) in fibroblast growth medium, reduced number of GcR (due to preincubation with very low density lipoproteins) led to a substantial increase in cholesterol synthesis. These findings indicate that the sensitivity of peripheral cells to glucocorticoids is decreased in HCh which might be one of the trigger mechanisms of atherogenesis.  相似文献

Extracts from the stems and leaves of Pitavia punctata Mol. were examined. The neutral fraction yielded β-sitosterol, daucosterin, quercetin, avicularin, and the previously undescribed quercetin 3-rhamnosylarabinoside. Braylin was co-extracted with the basic constituents, dictamnine, skimmianine and γ-fagarine. Acid hydrolysis of the leaves yielded cyanidin and delphinidin.  相似文献

Crosslinking of subunits of the high molecular weight oligomer of bovine heart mitochondrial creatine kinase (CKm) by dimethyl suberimidate and subsequent electrophoresis in the presence of sodium dodecyl sulfate gives eight protein bands. An increase in the time course of the enzyme crosslinking reaction results in the protein accumulation in the high molecular weight bands. Evidence has been obtained suggesting that crosslinking involves only the intraoligomeric contact areas. It is concluded that bovine heart CKm is an octamer. Crosslinking of intersubunit contacts in the octameric form of the enzyme by various diimidates has been carried out. The data obtained suggest that within the octamer the CKm subunits have a quasispherical rather than planar arrangement. This finding is supported by electron microscopy data.  相似文献
